Washu Fitzgerald Kobayashi, a 12-year-old American prodigy, arrives in Japan to become the new science teacher at Umi no Hoshi Elementary, but her real goal is to research the magical girl Pretty Sammy. However, her first day of teaching class 4-3 is interrupted by Pixy Misa and her Love-love Monster "Chemistry Girl".

On the lawn of the White House in Washington, D.C. Washu demonstrates a powerful new "GPS satellite camera" in space to the U.S. President and his assistant. However, as soon as she realizes that they only want to use it for military purposes, Washu activates a button that causes the satellite to self-destruct. She then receives a newspaper about a mysterious magical girl that has appeared in Tokyo. Washu decides that she wants to research this, and flies off to Japan using a rocket that she secretly built inside the White House while the president and his assistant were busy on their nightly dates.

At Sasami's school, Mihoshi announces that the school will be receiving a special instructor from America. Just then, Washu's rocket ship crashes into the school's courtyard and Washu emerges, proudly wearing the American Stars and Stripes as a cape before introducing herself as "Washu-chan". She then uses a high-tech fish-roll telescope to identify Sasami as the magical being.

In Juraihelm, Tsunami is watching the events at Umi no Hoshi Elementary, concluding that Washu must be one happy teacher, and turns to Ramia to get her opinion, but she's actually sleeping behind her on a short table. Tsunami then takes out a huge mallet to hit her with as punishment, but she changes her mind after Ramia mutters her name and she assumes that she's affectionately dreaming about her. She goes to fetch a cover for Ramia, but after she leaves, it's revealed that Ramia was actually dreaming about wanting to kill Tsunami.

After staring at the camera and announcing his name to the audience, Principal Miura asks Washu why, as a graduate of MIT at just 10 years old, she would want to teach at his school. She answers that she is on a journey to explore the depths of truth and that it's the nature of the solitary hunter. A clearly-confused Miura mumbles something about Sri Lanka before hiring Washu as a special science instructor. After Washu leaves, he then falls asleep on his desk before waking up, announcing that he fell asleep, and stating that his name is Principal Miura.

Washu kicks Mihoshi out of her own class 4-3 and announces that she came to Japan for the sole purpose of researching the magical girl Pretty Sammy, to Sasami's shock. She then walks over to Sasami and asks her where Sammy is. When she says she doesn't know, Washu explodes into anger and yells "Oh no!".

Rumiya, in his bird form, is watching the whole scene unfold from outside the school. He calls Ramia, who is still sleeping—now alongside Tsunami—what they should do for this week. Ramia simply tells him to "do it" and "do it right" before hanging up. Rumiya doesn't have much choice in the matter, though he doesn't know exactly what he's supposed to do.

Later, Washu takes over Mihoshi's class 4-3 for science instruction but is shocked to learn that the class's current science project is simply to extract starch from potatoes. Instead of doing such a simple experiment, she lets the class go wild with mixing chemicals that causes a foul aroma. Misao goes to open one of the windows and get some fresh air, but she ends up looking at Rumiya, who is holding a pendulum-swinging coin, and is hypnotized into going to the bathroom to secretly transform into Pixy Misa.

As Mihoshi notes that this wasn't the way she expected the class to go, Washu explains that she believes that the fastest way to meet Pretty Sammy is to cause a disturbance. Again, she goes over to Sasami and directly tells "Sammy" to come out. However, Pixy Misa runs into class. Washu tries to perform experiments on Misa once she learns that she's a magical girl, but Misa doesn't want to go along with them. She instead summons the Love-love Monster "Chemistry Girl", literally a giant, talking flask. Misa notices that Chemistry Girl doesn't have any feet, but Chemistry Girl insists that she doesn't need those "mere decorations". Then, on Pixy Misa's command, Chemistry Girl traps Washu inside a giant beaker that's slowly filling with potato starch. While this is supposed to be a torturous experience, Washu is instead fascinated with the data she's getting on her portable computer.

While everyone's distracted, Ryo-Ohki, who was hiding in one of the human dummies in the back of the class, bring out the magical baton for Sasami. She runs into the hallway and transforms into Pretty Sammy. When she runs back inside as Sammy, Pixy Misa breaks the fourth wall by suggesting that this is the end of Magical Girl Pretty Sammy, and the programming starting next will be "Pixy Misa and the Diary of her Gluttonous Journeys", though Sammy doubts that the show would be able to get any ratings.

Misa orders Chemistry Girl to attack Sammy, but as it turns out, without the "mere decorations" known as feet, Chemical Girl is unable to move. Already aware that she's lost, Misa then calmly walks out of the classroom and allows Sammy to easily defeat Chemical Girl with Pretty Coquettish Bomber. Washu is freed from the flask, but she drops her portable computer in the process, causing it to hit the floor and lose all of its data.

Ramia wakes up again and panics upon seeing the balance of Gemini shift toward good. She then calls Rumiya and angrily demands he to return to Juraihelm. Tsunami, still asleep, sits up for a moment and says "flowers... in a dream".

When Sasami returns home from school, she finds Washu eating soba noodles with her mother. As it turns out, Washu has moved into the house next door to hers. Later the night, Washu is playing the guitar just outside of Sasami's room. She asks if she wants to hear, but Sasami shuts the door. Washu continues to sing about how she'll never give up and find out about Sammy's secrets and the secret of magic.

Trivia

  • Washu's last name in this series, Kobayashi, is taken from her Japanese voice actor's name, Yuko Kobayashi
  • When Chemistry girl is created, she chants "H, He, Li, Be!". These are the symbols used for the first four elements of the Periodic Table: (H)ydrogen, (He)lium , (Li)thium, and (Be)ryllium.
  • During the scene where Konoha attempts to impress Hiroto with her knowledge of English, she either deliberately or accidentally says "Je t'aime", which is French for "I love you".
  • The newspaper Washu read in America that informed her of the existence of magical girls confirms that the show's events occur in Tokyo, Japan. Similarly, the OVA version of Pretty Sammy is also set in Tokyo, as evidenced by the second OVA's events taking place in Akihabara, a prominent commercial district in that city.
